Interaction Between Amorolfine and Voriconazole Against Fusarium species

چکیده
Fusarium species represent a range of fungal pathogens capable causing diverse mycotic diseases. Relative to antibacterial drugs, few effective antifungal agents have been developed date, and all are subject significant limitations. As such, there is an urgent need design novel treatments for infections caused by spp. Herein, 15 clinical isolates, including 5 oxysporum 10 solani strains, were analyzed explore the relative inhibitory effects different combinations amorolfine (AMO) voriconazole (VOR) on growth these pathogens. These analyses conducted measuring minimal concentration (MIC) values in broth microdilution assay using vivo model Fusarium-infected Galleria mellonella. experiments revealed that isolation, AMO VOR exhibited MIC ranging from 4 16 ?g/mL 2 8 ?g/mL, respectively. However, fell 1–2 0.5–2 respectively, when administered combination with one another, exhibiting synergistic activity against 73.3% strains. Subsequent G. mellonella further confirmed + treatment was associated significantly improved larval survival following infection. Together, results serve as first published evidence demonstrating exhibit spp., indicating they may approach disease treatment.

Synergysm of voriconazole or itraconazole with other antifungal agents against species of Fusarium.
BACKGROUND Infections caused by Fusarium are difficult to treat because these fungi show in vitro and in vivo resistance to practically all the antifungal agents available, which explains the high mortality rates. An attempt to overcome fungal resistance is the combination of antifungal agents, especially those with different mechanisms of action. AIMS Evaluate the in vitro interactions of co...
متن کاملMicrodilution susceptibility testing of amphotericin B, itraconazole, and voriconazole against clinical isolates of Aspergillus and Fusarium species.
We compared the activities of amphotericin B, itraconazole, and voriconazole against clinical Aspergillus (n = 82) and Fusarium (n = 22) isolates by a microdilution method adopted from the National Committee for Clinical Laboratory Standards (NCCLS-M27A). RPMI 1640 (RPMI), RPMI 1640 supplemented to 2% glucose (RPMI-2), and antibiotic medium 3 supplemented to 2% glucose (AM3) were used as test m...
متن کاملEffect of voriconazole combined with micafungin against Candida, Aspergillus, and Scedosporium spp. and Fusarium solani.
Effects of voriconazole combined with micafungin against 101 isolates of Candida spp. and 100 isolates of filamentous fungi have been evaluated by in vitro checkerboard analysis. The combination was indifferent for 97% of the Candida isolates and synergistic for 64% of the filamentous fungi (79% for Aspergillus fumigatus).

We compared the in vitro pharmacodynamics of amphotericin B, itraconazole, and voriconazole against Aspergillus, Fusarium, and Scedosporium species with a combination of two non-culture-based techniques: the tetrazolium salt 2,3-bis-(2-methoxy-4-nitro-5-[(sulfenylamino)carbonyl]-2H-tetrazolium-hydroxide) (XTT) colorimetric reduction assay, and fluorescent microscopy with the cellular morbidity ...
متن کاملIn vitro activity of voriconazole against Candida species.
The in vitro activity of voriconazole was compared with that of itraconazole and fluconazole against 181 isolates of Candida albicans, 124 isolates of Candida glabrata, and 20 isolates of Candida krusei obtained from the early 1980s through the mid-1990s. Voriconazole had greater intrinsic activity than fluconazole or itraconazole against all three Candida species. For C. glabrata, C. krusei, a...
